Abstract : Marine xylanolytic Bacillus subtilis APCMST-TS6 isolated from the marine sediments of Thengapattinum estuary was screened for the production of halophilic organic solvent tolerant xylanase (HOSX) by using halophilic production medium containing agricultural waste as a substrate. Purification and characterization of halophilic organic solvent tolerant xylanase produced by marine B. subtilis was performed through ammonium sulphate precipitation and anion exchange chromatography. The purified xylanase showed homogeny with 24.07 fold purity and 31.38% enzyme activity and it had the molecular mass of 37 kDa. This enzyme was characterized as alkaline thermophilic with optimum activity at pH 9 and 50ºC after 1.15hrs of incubation. This enzyme was also halophilic in nature and showed optimal activity at 1 M sodium choride. Metal ions such as magnesium sulfate, ammonium chlorideand magnesium chloride were enhanced this enzyme activity and its activity was also influenced by Tween 20. This enzyme was strongly inhibited by ethylenediaminetetraacetic acid (EDTA) and hence emerged as mettalo enzyme. The xylanase from B. subtilis registered maximum saccharification efficiency on the substrate such as pretreated wheat bran and rice straw. The properties of xylanase from B. subtilis confined its potential biotechnological applications.
