Abstract : Diabetes mellitus (DM) affects throughout body system including hepatobiliary system. Hepatocyte, an important hepatic parenchymal cell, is also disturbed. Consequently, the hepatic metabolism impairs and leading to severe hepatopathy. The aim of this study was to investigate the parameter changes of hepatocyte, nucleus, and nucleolus in diabetic rats by using morphometric analysis technique. Forty male adult Sprague-Dawley rats were used. Diabetic group (acute: n=10 and chronic: n=10 stages) were intraperitoneally injected with 60 mg/kg/BW of streptozotocin (STZ). Each control rats (acute: n=10 and chronic: n=10 stages) were injected with citrate buffer at pH 4.5 via the same route. Animals were sacrificed at acute (4 weeks) and chronic (24 weeks) periods. In diabetic group, whole blood glucose and urine glucose levels increased. Hepatic tissues were processed in traditional histological technique. Samples were examined under a light microscope and analyzed morphometric parameters of mononuclear hepatocyte in periportal (Z1) and pericentral (Z3) zones by using UTHSCSA Image tool (IT) 3.0 software. Mean areas of hepatocyte, nucleus, and nucleolus were increased in zone 1 and zone 3 of acute DM rats. Whereas, mean areas of hepatocyte, nucleus, and nucleolus were significantly reduced in both zones of chronic diabetic rats. Moreover, binuclear hepatocytes were increased in acute DM while they were significantly decreased in chronic diabetic rats. In conclusion, hepatic morphometric alterations occurred in DM rats that imitate diabetic patients. This data contributes the severity of diabetic hepatocytes with untreated insulin for further clinical research. Keywords - Morphometric Analysis, Hepatocyte, Diabetes Mellitus,
