Abstract : Diversity evaluation of the genetic resources is a prerequisite for the selection of suitable parents for variety
improvement. The objectives of this study were to assess the genetic diversity and relationships of 23 mungbean
(Vignaradiata(L.)Wilczek) and 4 blackgram (Vignamungo (L.) Hepper) genotypes, and to generate their molecular
fingerprints using expressed sequence tag (EST)-derived simple sequence repeat (EST-SSR) analysis. Fifty sixalleles were
amplified and high polymorphism (96.60%) was found from 11 EST-SSRs. The analysis found that number of alleles per
locus varied from 2 to 9, with an average of 5 alleles per locus while, values for the polymorphic information content (PIC)
varied from 0.22 to 0.80, with an average of 0.60 suggesting sufficient discriminatory power. Pair-wise coefficients of ESTSSR
based genetic similarity ranged from 0.61 to 1.00, with an average of 0.82. A dendrogram based on the UPGMA
analysis showed 2 main clusters; cluster I containing all mungbeanand cluster II containing all blackgram genotypes,
suggesting that EST-SSR is useful in the exploration of genetic diversity at the species level. Interestingly, mungbean
genotypes could be further sub-clustered with clear separation between genotypes reported to be resistant to diseases and the
ones having high yielding potential. These results indicate the usefulness of these mungbean and blackgram genotypes for
breeding purposes.
